The latest issue of Nintendo Power has confirmed that the rumors of a 3DS Castlevania game are true. Castlevania: Lords of Shadow - Mirror of Fate is currently in development at MercurySteam.
Fate will star Trevor Belmont. Trevor, if you recall, was one of the main characters of Castlevania III: Dracula's Curse. I don't expect any links between the plot of C3 and this game, though. The use of the subtitle Lords of Shadow implies that the game will follow the new storyline introduced in MercurySteam's PS3/Xbox 360 Lords from 2010.
Trevor fights the undead using his own version of the Combat Cross. He'll also employ secondary weapons like a glaive and electric bomb. The summary from GoNintendo notes that players can use experience points unlock combo moves that will allow Trevor to throw opponents in the air or slam them to the floor. The Light and Shadow magic system from Lords will make a return as well.
A few fans might be disappointed that this isn't a 2D "Metroidvania" game. Still, the writing was on the wall when it was revealed that MercurySteam is the developer. With Lords of Shadow, they established their own spin on the series. I don't think they would've been tapped for this 3DS project if Konami just wanted an old school Castlevania game.
There was no mention of a release window for Mirror of Fate. I'd imagine Konami will provide more details on the game sometime during E3 next week, though. It might even be part of Nintendo's press conference.
